چکیده
Currently, there are several treatments for osteoporosis however; they all display some sort of limitation and/or side effects making the need for new treatments imperative. We have previously demonstrated that NMP is a bioactive drug which enhances bone regeneration in vivo and acts as an enhancer of bone morphogenetic protein (BMP) in vitro. NMP also inhibits osteoclast differentiation and attenuates bone resorption. In the present study, we tested NMP as a bromodomain inhibitor and for osteoporosis prevention on ovariectomized (OVX) induced rats while treated systemically with NMP. Female Sprague-Dawley rats were ovariectomized and weekly NMP treatment was administrated 1 week after surgery for 15 weeks. Bone parameters and related serum biomarkers were analyzed. 15 weeks of NMP treatment decreased ovariectomy-induced gained weight in average by 43% and improved bone mineral density (BMD) and bone volume over total volume (BV/TV) in rat femur on average by 25% and 41% respectively. Moreover, mineral apposition rate and bone biomarkers of bone turnover in the treatment group were at similar levels with those of the Sham group. Due to the function of NMP as a low affinity bromodomain inhibitor and its mechanism of action involving osteoblasts/osteoclasts balance and inhibitory effect on inflammatory cytokines, NMP is a promising therapeutic compound for the prevention of osteoporosis.

Concentrations of N-methyl-2-pyrrolidone (NMP) and its metabolites in plasma and urine following oral administration of NMP to rats.
The primary aims were to study the metabolism in rats and to determine the biological levels after one oral developmentally toxic dose of N-methyl-2-pyrrolidone (NMP), a widely used industrial chemical. Non-pregnant female Sprague-Dawley rats were given an oral single dose of either a non-toxic dose of 125 mg NMP/kg (group 1) by gavage or a developmentally toxic dose of 500 mg/kg (group 2). Blo...
متن کاملVortex fluidic exfoliation of graphite and boron nitride.
Graphite is exfoliated into graphene by shearing vortex fluidic films of N-methyl-pyrrolidone (NMP), as a controlled process for preparing oxide free graphene, and for exfoliating the corresponding boron nitride sheets.
متن کاملRemoval N-Methyl Pyrrolidone from petrochemical effluent using photo-Fenton process: a case study of Mahshahr port petrochemical unit
Background and Objective: N-methylpyrrolidine is a highly potent solvent with toxic and adverse ecological properties that has historically been widely used in the chemical and petrochemical industries. Due to the health and safety، toxicity and environmental effects of effluents containing NMP، these effluents should be pre-treated before biological treatment methods. The aim of this study was...
متن کاملSonochemical degradation of N-methylpyrrolidone and its influence on single walled carbon nanotube dispersion.
Sonicating pure N-methyl pyrrolidone (NMP) rapidly produces contaminating organic nanoparticles, at increasing concentration with time, as investigated by AFM, as well as UV-vis, IR and NMR spectroscopies. The contamination issue affects carbon nanotube, and likely other nanomaterial, dispersions processed by sonication in organic solvents.
متن کاملLevels of N-methyl-2-pyrrolidone (NMP) and its metabolites in plasma and urine from volunteers after experimental exposure to NMP in dry and humid air.
The aim of this study was to investigate if the uptake of N-methyl-2-pyrrolidone (NMP), a widely used industrial chemical, increases after exposure to NMP in humid air compared to dry air. NMP has been described to be an airway irritant and a developmentally toxic compound. Six male volunteers were exposed to NMP, three at the time, for 8h in an exposure chamber. They were each exposed on four ...
